I've always heard it pronounced like 'gooey' as in "that cookie was gooey!"

It would be gooey on a touch screen if you ate peanut butter with your fingers first,but otherwise I think it's Gee-You-Eye.

GUI= Graphic User Interface. It's pronounced Gee-You-Aye.
